The 50C1 monoclonal antibody binds to human CD371, a 30 kDa type II transmembrane protein with extracellular C-type lectin domains, belonging to the C-type lectin family. CD371 is also known as CLEC12A (C-type lectin domain family 12, member A), C-type lectin-like molecule 1 (CLL-1), myeloid inhibitory C-type lectin-like receptor (MICL), and dendritic cell-associated lectin 2 (DCAL-2). It is expressed on monocytes, macrophages, dendritic cells, granulocytes, and some NK cell subsets. CD371 is structurally similar to the β-glucan receptor (dectin-1) and LOX-1 with high N-glycosylation. CD371 can be present in 5 distinct isoforms due to alternative splicing. CD371 downstream signaling can induce dendritic cell maturation and the production of IL-12. There is evidence that CD371 serves as a negative regulator of activated leukocytes recruited to sites of inflammation.
