The monoclonal antibody 7C9 recognizes Siglec-8, a 54 kDa type I transmembrane protein. Siglecs are cell surface receptors belonging to the immunoglobulin superfamily that recognize sugar antigens. Siglec-8 is specific for 6'-sulfo-sLex and is primarily expressed on eosinophils, with low-level expression in basophils and mast cells. The cytoplasmic domain of Siglec-8 has 2 immunoreceptor tyrosine-based inhibitor motifs (ITIM), which recruit SH2 family phosphatases following tyrosine phosphorylation. There is evidence Siglec-8 crosslinking decreases eosinophil viability through apoptotic induction.
