Terminal deoxynucleotidyl transferase (TdT) is a 60 kDa terminal tranferase, a specialized DNA polymerase that adds N nucleotides to the V, D, and J exons during antibody and T cell receptor gene recombination. TdT is expressed in the nucleus of pre-B and pre-T cells in addition to acute lymphoblastic leukemic cells. Mature lymphocytes from normal peripheral blood usually do not express the enzyme. TdT catalyzes the addition of nucleotides to the 3' terminus of a DNA molecule.
