Mcl-1 (myeloid cell leukemia-1) is an anti-apoptotic member of the Bcl-2 family found in mitochondrial and nonmitochondrial membranes. Mcl-1 dimerizes with Bax. Different from its other family members, Mcl-1 is highly regulated at both the transcriptional and post-translational level. Expression of Mcl-1 can be induced by various extracellular stimuli, including PMA, GM-CSF, M-CSF, IL-1β, IL-3, and IL-6 in hematopoietic cells. In response to growth factor stimulation, Mcl-1 undergoes transcriptional activation downstream of a number of well-known signal transduction pathways such as MAP kinases, PI3K/Akt, and JAK/STAT.
