p21 (CIP1/WAF1), also known as cyclin-dependent kinase inhibitor 1 or Cdk-interacting protein 1, binds to and inhibits cyclin-dependent kinase activity and thus blocks progression from G1 to S phase in the cell cycle. The expression of p21 is tightly controlled by the tumor suppressor protein p53. Upon DNA damage, p53 is phosphorylated and up-regulates p21 transcription via a p53-responsive element. p21 is expressed in all adult human tissues. The expression of p21 is also associated with favorable prognosis in various tumors.
