CD284, also known as Toll-like Receptor 4 (TLR4), is a 110 kDa member of the TLR family expressed on monocytes/macrophages, endothelial cells, and at low levels on B cells and granulocytes. Bacterial LPS, the major structural component of the outer wall of all gram-negative bacteria, engages TLR4 to induce a signaling cascade leading to the activation of an inflammatory response. TLR4 physically associates with MD-2, and together with CD14, this complex is responsible for LPS recognition and signaling. Binding of HTA125 to TLR4 is able to block LPS-induced cytokine expression.
