Epidermal growth factor receptor (EGFR) is a transmembrane tyrosine kinase belonging to the HER/ErbB protein family, and expressed by epithelial and endothelial cells. Ligand binding to EGFR results in receptor dimerization, autophosphorylation, activation of downstream signaling, internalization, and lysosomal degradation, leading to cell growth or differentiation. The Grb2 adapter protein binds activated EGFR at phospho-Tyr1068. EGFR has been shown to be frequently overexpressed or hyperactivated in a number of epithelial tumors. The downstream signaling effects of these aberrations lead to impaired apoptosis and/or enhanced proliferation, suggesting a causative relationship between receptor dysregulation and the pathology of many cancers. The D7A5 antibody reacts with EGFR phosphorylated at the tyrosine 1068 residue.
