An essential mechanism of cellular signal transduction is the phosphorylation of proteins at tyrosine, threonine, and serine residues. An increase in total cellular phosphotyrosine levels indicates a shift in the balance of tyrosine protein kinases and phosphases. Constitutively high total phosphotyrosine levels can be indicative of a dysregulated kinase, as in the case of the oncogenic BCR-ABL fusion protein.
