What is the difference between NO CALL and an Invalid call in the Fluidigm Genotyping Analysis software?
A NO CALL is assigned by the genotype-calling algorithm when, based on the confidence threshold set by the user, it cannot associate a given datapoint with a cluster. Although no genotype call has been assigned to the datapoint, a NO CALL remains part of the analyzed dataset. An Invalid call can only be manually assigned by a reviewer and should be used to remove datapoints from the analysis.
If data is reanalyzed after invalidating datapoints, the software will reanalyze the entire IFC, excluding the invalid datapoints. This may change automatic calls of datapoints, but any manually edited calls will not change upon re-analysis.
